The Kubicek laboratory at CeMM, the Research Center for Molecular Medicine of the Austrian Academy of Sciences in Vienna, is recruiting a synthetic organic chemist to generate novel small molecule chemical probes for chromatin biology.
The Position: The successful applicant will work as part of an ERC-funded team to develop a detailed understanding of the role of small molecule metabolites in the control of chromatin structure and gene expression. To do so, the candidate will apply her or his synthetic chemistry skills to design and generate novel small molecules that will be applied as chemical probes by closely collaborating biologists in the laboratory.
One focus on the project will be the generation of compounds that visualize and perturb the nuclear folate metabolism pathway, based on recent findings from the laboratory (https://www.biorxiv.org/content/early/2018/10/10/439422). The applicant will further work on the generalization of this compound synthesis strategy. The aim is to design novel chemical screening libraries by the modular assembly of compounds, enabling the systematic attachment of tags for fluorescent probing, chemical proteomics, and proteolytic degradation of protein targets.
The Candidate: The successful candidate holds a degree in Synthetic Organic Chemistry (Master or PhD degree), and has a proven track record in the application of synthetic pathways to systematically generate compound sets. Expertise in bioorthogonal chemistry, metabolomics or compound library design is considered a plus. The working language at CeMM is English, and excellent written and oral communication skills as well as high accuracy, reliability and excellent interpersonal and organizational skills are a requirement.
The Laboratory: The Kubicek laboratory is working on the role of chromatin in the definition of cell types and cell states, and the use of chemistry to modulate epigenetic states. The ERC-funded project “Chromabolism” focuses on a systematic understanding of the role of chromatin-bound central metabolic enzymes in the regulation of gene expression.
Recent publications include the first single-cell transcriptomes from primary human pancreatic islets (Li et. al EMBO Rep. 2016), the identification of artemisinins as compounds impacting alpha cell identity (Li et al. Cell 2017), novel functional screening for chromatin states (Sdelci et al. Nat. Chem. Biol. 2017) and synergistic drug combinations (Licciardello et al. Nat. Chem. Biol. 2017). Furthermore, the laboratory hosts the Platform Austria for Chemical Biology PLACEBO, a unique chemical screening infrastructure with a collection of 90,000 small molecule compounds. In a unique automated setup, the platform conducts up to 40 high-throughput screens in collaboration with internal and external researchers. In addition, we apply proteomics, metabolomics, chemistry, mass spectrometry, bioinformatics, and next generation sequencing to address our research questions. The selected candidate will have the opportunity to work in an innovative research environment and collaborate closely with other groups at CeMM.
