 Cloning of new giant virus genes into bacterial and amoebal plasmids.


You will participate in the Chaire d’Excellence project “Structural Biochemistry of Novel Metabolic     Pathways in Giant Viruses” led by Elsa Garcin and funded by A*MIDEX. This project focuses on the characterization of novel metabolic pathways in giant virusesYou will design and implement experimental protocols, and perform experiments under the supervision of Elsa Garcin and in collaboration with other team and lab members. You will learn new methodologies and techniques for this project. 

Activities :

  • Mutagenesis
  • Expression and production of recombinant proteins in bacteria and/or amoeba
  • If necessary, development of other expression systems (LEXSY, baculovirus, …)
  • Bacterial two-hybrid system
  • Purification of recombinant proteins for structural and interaction studies
  • Biochemical and biophysical characterization (SDS-PAGE, western blot, BLITZ, nano-DSF, DLS, ITC, …)
  • Work closely with undergraduates (L1-M2)


Your projects will be based on molecular biology, cellular biology, and protein biochemistry. You will express and purify proteins and complexes for structural studies (x-ray crystallography, SAXS, electron microscopy, NMR). Experience with heterologous expression in bacteria (i.e. Escherichia coli) is required. You will also work with the host of giant viruses, Acanthamoeba castellanii, and its associated viruses (Mimivirus, Megavirus, …). The project will be carried out in close collaboration with partner labs (IBS Grenoble, spectroscopy; BIP, EPR) and groups and platforms at the Institut de Microbiologie de la Mediterannée (EPR, Proteomics, Microscopy, Biophysics). 

Contract duration: maximum of 25-26 months (contract will end on December 31, 2024)

Salary: calculated based on the candidate’s experience


Education: Bac+5 (Master 2 or equivalent)

Experience: 0-1 years

The candidate should have knowledge and some experience in molecular biology, protein expression in bacteria (or other), protein purification. A background in basic techniques in molecular biology (PCR, cloning, site-directed mutagenesis), and/or biochemistry (protein expression and purification, SDS-PAGE, Western blot) is required. Experience in cellular biology is a plus but not required. We are looking for an enthusiastic, dynamic, organized person, who can work collaboratively with other team and lab members. Workplace language is French and/or English.

Special conditions: experiments with non-pathogenic bacteria (Escherichia coli) and on the host of giant viruses (Acanthamoeba castellanii), and its associated viruses (Mimivirus, Megavirus, …) will be necessary. For these, you will be required to strictly follow safety protocols established at the IGS. Some experiments may require to work at night or on weekends if necessary.
